Matreya LLC gd2 (cat. #1527)
Effect of aging on the rat brain GSs levels in individual rat brain structures. a–i Changes in GSs levels ( a GM1, b GD1a, c GT1b, d GD3, e GD1b, f GQ1b, g <t>GD2,</t> h GM3, i GM2); ordered by abundance) expressed as µg of GSs per mg of brain tissue DW. The levels were determined in 1 month and 11 months aged groups in the neocortex (NEO), allocortex (ALLO), cerebellum (CB), olfactory bulbs (OB), medial basal forebrain (MF), hypothalamus (HYPO), brainstem (BS), cingulate cortex (CING), striatum (STR), ventral (VHIP) and dorsal hippocampus (DHIP). Data were analyzed using an adjusted F test with Kenward-Roger type adjustment of denominator degrees of freedom. n = 13 (NEO 12) for the group of 1-month-old rats and n = 13 (DHIP 12) for 11 months old rats. Statistical significance is indicated with *p ≤ 0.05, **p ≤ 0.01, ***p ≤ 0.001. The boxes span from the 25th to the 75th percentiles, the lines in the middle denote the median values, and the whiskers extend from the smallest values and up to the largest values (dots show all data points).

Techniques:

Bar chart depicting the a-series to b-series GSs in the brain regions. The a-series GS included GM1, GM2, GD1a, and GM3, whereas the b-series included GD1b, GT1b, GQ1b, GD2, and GD3. Statistical significance is indicated with *p ≤ 0.05. The regions with the starkest changes during neurodevelopment were the hippocampus, hypothalamus, medial forebrain, and brainstem.

Techniques:

Cell fluorescence imaging and CCK-8 assay of the novel fluorescent molecule probe. (A) Labeled hUC-MSC suspension exhibited moderate fluorescence on small animal fluorescent imager, whereas none was detected in the unmarked hUC-MSC suspension. The hUC-MSCs were efficiently labeled by Cy7-GD. (B) CCK-8 assay to evaluate the cytotoxicity of the Cy7-GD2 complexes. No statistically significant difference was found between the two groups (P>0.05). The results are presented as the mean ± standard deviation (n=5 per group). hUC-MSCs, human umbilical cord-derived mesenchymal stem cells; CCK-8, Cell Counting Kit-8; GD2, ganglioside; Cy7, CyDye mono-reactive NHS esters.
